There's no rule says you have to.
Typically, challenging for a world title signifies you're ready to step up in class (or at least, someone thinks you're ready). Obviously some come straight back with a crash but there is no actual BBBoC rule that says you must vacate.

Chisora is under no obligation to defend either British or Commonwealth titles for another 9 months if he doesn't want to. Sexton was the mandatory for the British and Dereck was for the Commonwealth he won from Sexton.

The EBU have one but it is caveated, the BBBoC (who govern British, English, Area and Commonwealth title fights) don't.

sorry to be pedantic but the Commonwealth Council govern the Commonwealth title, not the Board. Good point. Threw me because the bbboc keep mentioning the Commonwealth in their rules.
